Divastin E 20 mg+10 mg (Tablet)

Indications

• Reduction of elevated total cholesterol
• Reduction of LDL-C levels
• Reduction of triglycerides
• Increase of HDL-C levels
• Adjunct to diet for hypercholesterolemia
• Treatment for homozygous familial hypercholesterolemia
• Reduction of hepatic cholesterol stores

Pharmacology

• Inhibition of HMG-CoA reductase
• Reduction of LDL production
• Increase of hepatic LDL receptors
• Inhibition of cholesterol absorption by the small intestine
• Targeting the sterol transporter NPC1L1
• Localizing at the brush border of the small intestine

Dosage & Administration

• Recommended starting dose of 10/10 mg or 20/10 mg daily
• Starting dose for patients with greater than 55% reduction in LDL-C
• Dosage range of 10/10 mg to 80/20 mg daily
• Monitoring lipid levels within 2 weeks of initiation
• Dosage adjustment based on lipid levels
• Dosage for homozygous familial hypercholesterolemia

Interaction

• Increased risk of myopathy with concurrent use of certain medications
• Avoid coadministration with cyclosporine or gemfibrozil
• Enhanced risk when used with niacin

Contraindications

• Active liver disease
• Hypersensitivity to any component

Side Effects

• Rhabdomyolysis
• Myopathy
• Liver enzyme abnormalities
• Myalgia
• Abdominal pain

Pregnancy & Lactation

• Contraindicated in pregnancy
• Increased serum cholesterol and triglycerides during pregnancy
• Unknown excretion in human milk

Precautions & Warnings

• Temporary withdrawal in case of myopathy
• Liver enzyme tests prior to initiating therapy
• Caution with drugs that may decrease endogenous steroid hormones

Use in Special Populations

• Safety and effectiveness not established in pediatric patients
