Indications

  • Treatment of chronic iron overload

Pharmacology

  • Orally active chelator
  • Highly selective for iron
  • Tridentate ligand
  • Excretion of iron in faeces
  • Low affinity for zinc and copper
  • Iron balance metabolic study

Dosage

  • Initial daily dose of 20-40 mg/kg body weight
  • Serum ferritin monitoring
  • Dose adjustment based on trends in serum ferritin
  • Dispersion in water or fruit juice
  • Empty stomach administration

Administration

  • Once daily on an empty stomach
  • Dispersion in water or fruit juice
  • Restrictions on consuming carbonated drinks or milk

Interaction

  • Avoidance with aluminium-containing antacids

Contraindications

  • Gastrointestinal bleeding, ulceration, and irritation
  • Cytopenias monitoring
