Description
- A synthetic, antibacterial agent with in vitro activity against Gram positive aerobic bacteria, some Gram positive anaerobic bacteria, and certain Gram negative bacteria.
- Selectively inhibits bacterial protein synthesis via a mechanism of action different from that of other antibacterial agents.
- Available in parenteral formulation for intravenous administration and oral presentation in the form of film-coated tablets and oral suspension.
Indications
- Vancomycin-Resistant Enterococcus faecium infections
- Nosocomial pneumonia caused by Staphylococcus aureus and Streptococcus pneumoniae
- Complicated skin and skin structure infections including diabetic foot infections
- Community-acquired pneumonia
- Bacteremia
Pharmacology
- Binds to the 23S ribosomal RNA of the 50S subunit of the bacterial ribosome
- Prevents the formation of a functional 70S initiation complex
- Bacteriostatic against enterococci and staphylococci
- Bactericidal for the majority of streptococci strains

- Oral bioavailability of approximately 100%
- Injection administered over a period of 30 to 120 minutes

Contraindications
- Known hypersensitivity to Linezolid or any of the product components
- Concurrent use of monoamine oxidase inhibitors
- Uncontrolled hypertension, pheochromocytoma, thyrotoxicosis, carcinoid syndrome
- Use with sympathomimetic, vasopressive, dopaminergic agents, serotonin re-uptake inhibitors, tricyclic antidepressants, and other specific medications
Side Effects
- Mild to moderate adverse events
- Common events include diarrhea, headache, and nausea
- Other events such as oral and vaginal moniliasis, hypertension, dyspepsia, localized abdominal pain, pruritus, and tongue discoloration
Pregnancy & Lactation
- Pregnancy Category C
- Caution advised during lactation due to potential excretion in human milk
Precautions & Warnings
- Monitoring for recurrent nausea or vomiting, unexplained acidosis, and low bicarbonate level
- Observation for signs and symptoms of serotonin syndrome
- Prompt ophthalmic evaluation if visual impairment symptoms occur
- Risk of convulsions in some cases with a history of seizures or risk factors
