Description

  • A sleep-inducing agent with rapid onset and short duration of action.
  • Exerts anxiolytic, anticonvulsant, and muscle-relaxant effects.

Indications

  • Short-term treatment of insomnia
  • Sedation in premedication before surgical or diagnostic procedures

Mode of Action

  • Acts through the inhibitory neurotransmitter gamma-aminobutyric acid (GABA)
  • Increases the activity of GABA to produce sedating, muscle-relaxing, and sleep-inducing effects
  • Binds to the benzodiazepine site on GABA-A receptors

Dosage & Administration

  • Oral dosage for adults: 7.5-15 mg daily
  • In premedication: 15 mg given 30-60 minutes before the procedure
  • Intravenous administration for endoscopic or cardiovascular procedures: Initial dose approximately 2.5 mg for healthy adults
  • Intramuscular administration in adults: 0.07-0.1 mg/kg body weight

Interaction

  • Can enhance the central sedative effect of various drugs including neuroleptics, analgesics, and antiepileptic drugs

Contraindications

  • Severe respiratory insufficiency
  • Severe hepatic insufficiency
  • Myasthenia gravis
  • Sleep apnea syndrome
  • Known hypersensitivity to benzodiazepines or any component of the product

Side Effects

  • Drowsiness, confusion, fatigue, headache, and muscle weakness may occur at the start of therapy and usually disappear with repeated administration
  • Fluctuations in vital signs including respiratory depression and variations in blood pressure and pulse rate

Pregnancy & Lactation

  • Avoid during pregnancy unless there is no safer alternative
  • Should not be administered to breastfeeding mothers

Precautions & Warnings

  • Dormax IV should be administered very slowly

Overdose Effects

  • Extreme overdosage may lead to coma, areflexia, and cardiorespiratory depression
  • Effects of overdosage can be controlled with benzodiazepine antagonist flumazenil
