Indications
- Short-term treatment of insomnia
- Sedation in premedication before surgical or diagnostic procedures
Description
Sleep-inducing agent with rapid onset and short duration of action. Also exhibits anxiolytic, anticonvulsant, and muscle-relaxant effects.
Pharmacology
Binds to GABA-A receptors to increase the activity of GABA, producing a sedating effect, relaxing skeletal muscles, and inducing sleep, anesthesia, and amnesia.
Dosage & Administration
- Oral dosage:
- 7.5-15 mg daily for adults
- 7.5 mg for elderly and debilitated patients
- 15 mg for premedication
- Intravenous administration:
- 2.5 mg for endoscopic or cardiovascular procedures in healthy adults
- 10-15 mg for induction of anesthesia
- 0.07-0.1 mg/kg body weight for adults via intramuscular administration
- Rectal administration in children:
- 0.35-0.45 mg/kg for preoperative sedation
Interaction
Can enhance the central sedative effect of various drugs including neuroleptics, antidepressants, analgesics, anesthetics, antipsychotics, and sedative antihistamines.
Contraindications
Not recommended for patients with severe respiratory or hepatic insufficiency, myasthenia gravis, sleep apnea syndrome, or known hypersensitivity to benzodiazepines.
Side Effects
- Drowsiness, confusion, fatigue, headache, muscle weakness
- Respiratory depression, apnea, variations in blood pressure and pulse rate after parenteral administration
Pregnancy & Lactation
Avoid during pregnancy and breastfeeding unless no safer alternative is available.
Precautions & Warnings
Dormitol IV should be administered very slowly.
Overdose Effects
Extreme overdosage may lead to coma, areflexia, cardiorespiratory depression, and apnea. Can be controlled with benzodiazepine antagonist flumazenil.
