Indications
- Short-term treatment of insomnia
- Sedation in premedication before surgical or diagnostic procedures
Description
Sleep-inducing agent with rapid onset and short duration of action. Also exerts anxiolytic, anticonvulsant, and muscle-relaxant effects
Pharmacology
Mediated through the inhibitory neurotransmitter gamma-aminobutyric acid (GABA). Increases GABA activity producing sedation, muscle relaxation, and inducing sleep, anesthesia, and amnesia.
Dosage & Administration
- Oral dosage for adults: 7.5-15 mg daily
- Intravenous administration for endoscopic or cardiovascular procedures: Initial dose approximately 2.5 mg for healthy adults
- Intramuscular administration for adults: 0.07-0.1 mg/kg body weight
- Rectal administration in children for preoperative sedation: 0.35-0.45 mg/kg
Interaction
Enhances the central sedative effect of various drugs including neuroleptics, antidepressants, analgesics, and antiepileptic drugs
Contraindications
Must not be given to patients with severe respiratory insufficiency, severe hepatic insufficiency, myasthenia gravis, sleep apnea syndrome, and known hypersensitivity to benzodiazepines

Pregnancy & Lactation

Precautions & Warnings
Administer IV very slowly
Overdose Effects
May lead to coma, areflexia, cardiorespiratory depression, and apnea. Can be controlled with benzodiazepine antagonist flumazenil

Storage Conditions
Protect from light and moisture, store in cool and dry place. Keep out of the reach of children
