Pharmacology
- Attaches to specific site on the GABA receptor
- Potentiates the effect of GABA
- Rapid absorption after oral administration
- Complete absorption after oral administration
- Long plasma half-life

- Metabolized in the liver
- Minimal unchanged drug excreted in urine
- Metabolites excreted through the bile into intestine
Dosage & Administration
- Oral administration for anxiety: 2 mg thrice daily
- Oral administration for insomnia associated with anxiety: 5-15 mg at bedtime
- Oral administration for night terrors and somnambulism in children: 1-5 mg at bedtime
- IM/slow IV injection for severe acute anxiety: 10 mg repeated if necessary
- IM/slow IV injection for febrile convulsion in children: Slow IV in a dose of 250 mcg/kg
- Rectal administration: A dose of 500 mcg/kg for children
Interaction
- Contraindicated with alcohol intake
- Increased sedation with concomitant use of neuroleptics, hypnotics, sedative antihistamines and CNS depressants
- Increased clearance when administered with phonobarbitone
- Decreased clearance when administered with cimetidine
- Inhibition of metabolism by omeprazole and isoniazide
