- Benzodiazepine derivative

Mode of Action
- Attaches to specific site on the GABA receptor
- Potentiates the effect of GABA
- Opens chloride ion channels into cells
- Rapid and complete absorption after oral administration
- Metabolized in the liver
- Peak plasma concentration reaches within 15-90 minutes
- Mean plasma half-life is 30 hours
- Plasma protein binding is 98-99%
- Small proportion of metabolites excreted through bile into the intestine and eliminated with the feces
Dosage and Administration
- Oral administration for anxiety, insomnia associated with anxiety, and night terrors in children
- IM/slow IV injection for severe acute anxiety, control of panic attacks, acute alcohol withdrawal, and febrile convulsion in children
- Rectal administration for children
Interaction
- Contraindicated with alcohol
- Increased sedation with concomitant use of neuroleptics, hypnotics, sedative antihistamines, and CNS depressants
- Pharmapam clearance increased by phonobarbitone and decreased by cimetidine
- Inhibition of Pharmapam metabolism by omeprazole and isoniazide
