Pharmacology
- Attaches to specific site on GABA receptor
- Potentiates effect of GABA
- Opens chloride ion channels into cells
- Rapid and complete absorption after oral administration
- Peak plasma concentration in 15-90 minutes
- Mean plasma half-life of 30 hours
- Metabolized in the liver
- Traces of unchanged drug excreted in urine
- Metabolites excreted through bile into the intestine
- Eliminated with the feces
- Significant absorption after rectal administration
- Peak concentration within 1.5-2 hours
Dosage & Administration
- Oral administration for anxiety
- Oral administration for insomnia associated with anxiety
- Oral administration for night terrors and somnambulism in children
- IM/slow IV injection for severe acute anxiety and acute alcohol withdrawal
- IM/slow IV injection for febrile convulsion in children
- Rectal administration in case of children
Interaction
- Concomitant intake with alcohol not recommended
- Increased sedation due to concomitant use of neuroleptics, hypnotics, sedative antihistamines, and CNS depressants
- Relaxen clearance increased by concomitant administration of phenobarbitone
- Relaxen clearance decreased by administration of cimetidine
- Inhibition of Relaxen metabolism by omeprazole and isoniazid
Contraindications
- Myasthenia gravis
- Pulmonary insufficiency
- Respiratory depression
- Hypersensitivity to benzodiazepine
