Pharmacology
- Attaches to specific site on GABA receptor
- Potentiates the effect of GABA
- Rapid and complete absorption after oral administration
- Peak plasma concentration within 15-90 minutes
- Mean plasma half-life of 30 hours
- Metabolized in the liver
- Metabolites excreted in urine and through the bile into the intestine
- Eliminated with the feces
- Significantly absorbed after rectal administration
- Peak concentration reaches within 1.5-2 hours
Dosage & Administration
- Oral administration for anxiety, insomnia, night terrors, and somnambulism
- IM/slow IV injection for severe acute anxiety, acute panic attacks, acute alcohol withdrawal, and febrile convulsion in children
- Rectal administration for children
- Different doses for specific conditions
Interaction
- Contraindicated with alcohol
- Increased sedation with concomitant use of neuroleptics, hypnotics, sedative antihistamines, and CNS depressants
- Clearance increased by concomitant administration of phenobarbitone
- Clearance decreased by administration of cimetidine
- Inhibition of metabolism by omeprazole and isoniazide
