- Benzodiazepine derivative

Pharmacology
- Attaches to specific site on the GABA receptor
- Potentiates the effect of GABA
- Opens chloride ion channels into cells
- Rapid and complete absorption after oral administration
- Peak plasma concentration within 15-90 minutes
- Mean plasma half-life of 30 hours
- 98-99% plasma protein binding
- Metabolized in the liver
- Traces of unchanged drug excreted in urine
- Metabolites excreted through the bile into the intestine
- Eliminated with the feces
- Significantly absorbed after rectal administration in suppository form
- Peak concentration reaches within 1.5-2 hours
Dosage & Administration
- Oral administration for anxiety relief
- Oral dosage 2 mg thrice daily
- Increased dosage to 15-30 mg daily if necessary
- Elderly or debilitated patients to receive half of the adult dose
- Insomnia treatment dosage 5-15 mg at bedtime
- Night terrors and somnambulism treatment dosage 1-5 mg at bedtime
- IM/slow IV injection for severe acute anxiety
- Control of acute panic attacks with IM/slow IV injection
- Large vein, rate below 5 mg/minute for slow IV injection
- Dosage for febrile convulsion in children
- Rectal administration in case of children
- Rectal dosage of 500 mcg/kg (maximum 10 mg)
Interaction
- Concomitant intake with alcohol not recommended
- Increased sedation due to concomitant use of neuroleptics, hypnotics, sedative antihistamines, and CNS depressants
- Seequil-S clearance increased by concomitant administration of phonobarbitone
- Seequil-S clearance decreased by administration of cimetidine
- Inhibition of Seequil-S metabolism by omeprazole and isoniazide

- Myasthenia gravis
- Pulmonary insufficiency
- Respiratory depression
- Hypersensitivity to benzodiazepine
