General Dosage

Dosage for Anxiety States
- Initial dose usually 20mg daily for adults
- Maximum daily dose of 30mg for adults
- Low initial doses for the elderly and children
- Secondary dosage adjustment after symptom improvement
Dosage for Epilepsy
- Small initial doses for adults and children
- Monitoring and re-assessment of patient's need for continued treatment
Administration
- Tablets can be administered whole or crushed and mixed in applesauce
- Can be taken with or without food
Interaction
- Increased bioavailability with alcohol consumption
- Potentiating effect with CNS depressant drugs
- Risk of sedation and respiratory depression with opioids
- Dosage adjustment when administered with anticonvulsants
- Enhanced euphoria when used concomitantly with narcotic analgesics
Contraindications
- Hypersensitivity to clobazam or excipients
- Myasthenia gravis
- Severe respiratory insufciency
- Sleep apnoea syndrome
- Severe impairment of liver function
- Breastfeeding
- Children between 6 months and 3 years
Side Effects
- Common: decreased appetite, irritability, somnolence, sedation, dry mouth, fatigue
- Uncommon: diplopia, muscle spasms, rash
- Not known: respiratory depression, photosensitivity reaction, muscle weakness
Pregnancy & Lactation
- Not recommended during first trimester of pregnancy
- Passes into breast milk, must not be used in breastfeeding women
Precautions & Warnings
- Closely monitor for signs or symptoms of serious skin reactions
- Risk of respiratory depression, muscle weakness, and fall in elderly patients
- Consideration of decreased anticonvulsant efficacy in the course of treatment
- Potential increased exposure in CYP2C19 poor metabolizers
- Suicidality risk and need for monitoring
- Interactions with CYP2C19 inhibitors
