Mode of Action
- Neuroleptic with anxiolytic and antidepressant properties
- Bipolar thymoleptic with activating properties
- Maximal serum concentration reached in about 4 hours after oral administration
- Biological half-life of about 35 hours
- Does not influence the pharmacokinetic properties of the individual compounds
Dosage and Administration
- Usually 2 tablets orally daily in the morning and noon for adults
- 1 tablet in the morning for elderly patients
- Maintenance dose usually 1 tablet orally in the morning
- Additional treatment with a sedative in the acute phase recommended for insomnia or severe restlessness
Interaction
- Enhances response to alcohol and other CNS depressants
- Simultaneous administration of MAO-inhibitors may cause hypertensive crises
- Reduces the antihypertensive effect of guanethidine and similar acting compounds
- Enhances the effects of adrenaline and noradrenaline
Contraindications
- Immediate recovery phase after myocardial infarction
- Defects in bundle-branch conduction
- Untreated narrow-angle glaucoma
- Acute alcohol, barbiturate and opiate intoxications
- Patients who have received an MAO-inhibitor within two weeks
- Not recommended for excitable or overactive patients
Side Effects
- Rare in recommended doses
- Transient restlessness
- Insomnia
Pregnancy and Lactation
- Preferably not given during pregnancy and lactation
Precautions and Warnings
- Withdrawal of previous tranquillizers with sedative effect gradually recommended

- Symptomatic and Supportive treatment
- Gastric lavage and administration of activated charcoal
- Measures aimed at supporting the respiratory and cardiovascular systems
- Use of diazepam for convulsions and biperiden for extrapyramidal symptoms
