Dosage
- Initial dose 5-10 mg once daily
- Subsequent dosage adjustments based on clinical status

Administration
- Can be given without regard to meals
- Gradual tapering of the dose recommended while discontinuing
Interaction
- May antagonize effects of levodopa and dopamine agonists
- Inducers of CYP1A2 or glucoronyl transferase enzymes may increase clearance
- Inhibitors of CYP1A2 may inhibit elimination
- Carbamazepine may increase clearance
- Activated charcoal reduces oral bioavailability by 50-60%
- Caution with centrally acting drugs and alcohol
Contraindications
- Hypersensitivity to any ingredient
- Known risk for narrow-angle glaucoma
Side Effects
- Very common: somnolence and weight gain
- Other effects: increased appetite, elevated glucose levels, elevated triglyceride levels, dizziness, akathisia, Parkinson's disease, dyskinesia, orthostatic hypotension, anticholinergic effects, asthenia, edema, photosensitivity reaction
Pregnancy & Lactation
- Use in pregnancy if potential benefits justify potential risk to the foetus
- Patients advised to notify physician if pregnant or intending to become pregnant during treatment
- No reported teratogenicity
- Patients advised not to breastfeed while taking Olanzapine
Precautions & Warnings
- Caution in patients with history of seizures
- Caution in patients with low leukocyte and/or neutrophil counts for any reason
- Caution in patients with impaired hepatic function
- Caution in patients using hepatotoxic medicines
- Caution in patients using centrally acting drugs
- Caution in patients using medicines known to increase QT interval
- Caution in elderly patients operating hazardous machinery or vehicles
Use in Special Populations
- Not studied in subjects under 18 years of age
- Starting dose for elderly patients (age 65 and over): 5 mg/day
- Starting dose for patients with hepatic or renal impairment: 5 mg/day
