Indications
- Treatment of acute and chronic psychoses
- Treatment of schizophrenia
- Treatment of bipolar disorder including manic episodes and depressive episodes
- Maintenance treatment of bipolar I disorder
Pharmacology
- Belongs to dibenzothiazepine derivatives
- Antagonist at multiple neurotransmitter receptors
- No appreciable affinity at cholinergic muscarinic and benzodiazepine receptors
- Mechanism of action is unknown
- Efficacy in schizophrenia is mediated through dopamine D2 and serotonin 5HT2 antagonism
Dosage & Administration
- Administered twice daily
- Total daily dose for the first four days of therapy
- Dosage adjustments based on clinical response and tolerability
- Usual effective dose range
- Administered once daily at bedtime for depressive episodes associated with bipolar disorder
Interaction
- Caution with medicines causing electrolyte imbalance or increasing QT interval
- Co-administration with thioridazine, carbamazepine, phenytoin increased Qmax clearance
Contraindications
- Hypersensitivity to Quetiapine

- Safety and efficacy not established during human pregnancy

- Breastfeeding while taking Quetiapine should be avoided
Precautions & Warnings
- Increased risk of suicidal thoughts and self-harm
- Caution in patients with cardiovascular or cerebrovascular disease
- Caution in patients with a history of seizures
- Consideration of dose reduction or discontinuation if tardive dyskinesia appears
- Discontinuation and medical treatment for neuroleptic malignant syndrome
- Caution in patients with cardiovascular disease or family history of QT prolongation
- Monitoring for severe neutropenia and hyperglycemia
- Elderly use with caution
- Not indicated for use in children and adolescents below 18 years of age

Overdose Effects
- Survival reported in acute overdoses
