Indications
- Treatment of acute and chronic psychoses
- Treatment of mania
- Treatment of schizophrenia
Pharmacology
- Selective monoaminergic antagonist
- High affinity for serotoninergic 5-HT2 and dopaminergic D2 receptors
- Binds to alpha1 adrenergic receptors
- Binds with lower affinity to H1 histamine and alpha2 adrenergic receptors
- Potent D2 antagonist
- Improves positive symptoms of schizophrenia
- Less depression of motor activity
- Reduces extrapyramidal side effect liability
- Extends therapeutic activity to negative and affective symptoms of schizophrenia
Dosage & Administration
- Initial dose of 2 mg in 1-2 divided doses on 1st day for psychoses
- Usual dose range of 4-6 mg daily for psychoses
- Initial dose of 2 mg once daily for mania
- Usual dose range of 1-6 mg daily for mania
- Risperidone generally administered at 1 mg BID initially for schizophrenia
- Target dose of 3 mg BID for schizophrenia
- Elderly or in hepatic or renal impairment: initial dose of 1 mg daily in two divided doses
Interaction
- May antagonize the effects of levodopa and dopamine agonists
- Chronic administration with Carbamazepine reduces plasma clearance of the product
- Chronic administration with Clozapine may decrease the clearance of the product
- May enhance the effects of certain antihypertensives
