Indications
- Acute and chronic psychoses
- Mania
- Schizophrenia
Pharmacology
- Selective monoaminergic antagonist
- High affinity for serotoninergic 5-HT2 and dopaminergic D2 receptors
- Binds to alpha1 adrenergic receptors
- Low affinity for H1 histamine and alpha2 adrenergic receptors
- Balanced central serotonin and dopamine antagonism
Dosage & Administration
- Initial dose of 2 mg in 1-2 divided doses
- Usual dose range of 4-6 mg daily
- Maximum dose of 16 mg daily
- Titration for elderly or hepatic/renal impairment
- Not recommended for children under 15 years
- Initial dose for mania and schizophrenia
- Dosage adjustments at intervals of not less than 1 week
Interaction
- May antagonize effects of levodopa and dopamine agonists
- Reduced plasma clearance with chronic administration of Carbamazepine
- Decreased clearance with chronic administration of Clozapine
- Enhancement of effects of certain antihypertensives
