Indications
- Major Depressive Disorder

Pharmacology
- Potentiation of serotonergic activity
- Inhibition of neuronal reuptake of serotonin
- Blockage of serotonin uptake into human platelets
- Selective inhibitor of neuronal serotonin reuptake
- Negligible effects on norepinephrine and dopamine reuptake
Dosage & Administration
- Usual Initial Dosage: 20 mg/day
- Maximum Dosage: 50 mg/day
- Maintenance Therapy: efficacy is maintained for periods of up to 1 year
Interaction
- Absorption not affected by food or antacids
- Potential interaction with Tryptophan
- Metabolism affected by drug metabolizing enzyme inducers/inhibitors
- Contraindicated with MAOIs and thioridazine
- Caution advised with alcohol, haloperidol, MAOIs, lithium, phenytoin, anticonvulsants, and warfarin

Precautions & Warnings
- Caution advised in cardiac conditions
- Caution in patients with epilepsy
- Discontinue in case of seizures
- Little clinical experience of concurrent administration with ECT
- No impairment of cognitive or psychomotor function observed
- Taper phase regimen for discontinuation of treatment
Use in Special Populations
- Recommended initial dose: 10 mg/day for elderly, debilitated, and/or severe renal or hepatic impairment
- Dosage should not exceed 40 mg/day
