Indications

Description
- Tranquilizer of phenothiazine type
- Tricyclic antidepressant with less sedative action
- Effective for anxiety and depression
- Restores functional ability without drug dependence
Pharmacology
- Nortriptyline inhibits uptake of norepinephrine and serotonin
- Fluphenazine acts as a neuroleptic drug with dopamine receptor antagonism
- Nortriptyline has greater effect on norepinephrine reuptake than serotonin reuptake
Dosage & Administration
- One tablet 2 to 3 times daily for adults
Interaction
- Interactions with barbiturates, alcohol, and narcotic drugs
- Central depressants should be administered with caution
Contraindications
- History of gradual epilepsy or organic brain damage
- Blood dyscrasia
- Severe cardiac insufficiency
- Renal or liver damage
- Patients taking monoamine oxidase inhibitor (MAOI)
- Younger children
- Hypersensitivity to any component of the preparation
Side Effects
- Tardive dyskinesias reported in phenothiazine therapy
- Blood dyscrasia
- Malignant Neuroleptic syndrome
- Sudden death reported rarely
- Agranulocytosis as a rare but potentially fatal adverse effect
Pregnancy & Lactation
- Safety in human pregnancy not established
- Not recommended for use in lactation as it is excreted in breast milk
Precautions & Warnings
- Precautions in patients with glaucoma, prostate enlargement, cardiac failure, and myocardial infarction
- Concurrent administration with CNS depressants
- May impair alertness and ability to drive or operate machinery
