Description
- Actions in the treatment of peripheral vascular disease
- Anti-vasoconstrictor properties
- Action on blood hyperviscosity
- Anti-ischaemic effect
- Selective calcium antagonist
- Inhibition of the influx of Ca2+ intracellularly
- Prevention of calcium entry into ischaemic erythrocytes
Pharmacology
- Antihistamine
- Labyrinthine sedative
- Peripheral antivasoconstrictor
- Inhibiting the influx of Ca2+ intracellularly
- Preventing the Ca2+ dependent contraction of arterial smooth muscle
- Improves vestibular symptoms
- Prevents peripheral arterial disease
Dosage & Administration
- Usual adult dose of 15 to 30 mg three times daily
- Children's dose is half of the adult dose
- Motion sickness dose of 30 mg two hours before the journey and 15 mg every 8 hours during the journey
- Peripheral arterial diseases dose of 75 mg two or three times daily
Interaction
- No drug interactions seen with antihypertensives
- No drug interactions seen with diuretics
- No drug interactions seen with anticoagulants
- No drug interactions seen with hypoglycaemics
Contraindications
- Decreases blood pressure significantly
- Reasonable caution in hypotensive patients
Side Effects
- Transient somnolence
- Transient gastrointestinal disturbances
- Allergic skin reactions on rare occasions
- Fatigue on rare occasions
- Aggravation or appearance of extrapyramidal symptoms in elderly people during prolonged therapy
Pregnancy & Lactation
- Safety not established in human pregnancy
- Animal studies show no teratogenic effects
- Not advisable to administer in pregnancy
Precautions & Warnings
- May cause drowsiness
- Patients affected should not drive or operate machinery
- Avoid alcoholic drink
