Indications
- Prevention and treatment of nausea

Pharmacology
- Meclizine is a piperazine-derivative antihistamine
- It has antiemetic, anticholinergic and antihistaminic properties
- Reduces the sensitivity of the labyrinthine apparatus
- Pyridoxine is vitamin B-6
- Pyridoxine has been added to enhance the anti-emetic effects
- Pyridoxine is added as a dietary supplement
Dosage & Administration
- 25-50 mg daily for adults and children 12 years of age & over for nausea and vomiting
- Initial dose of 25-50 mg for motion sickness, to be repeated every 24 hours if needed
- 50 mg for radiation sickness administered 2-12 hours prior to treatment
- 25-100 mg daily in divided doses for vertigo
- 25-50 mg for prevention of nausea and vomiting associated with emergency contraceptive pill (ECP)
Interaction
- CNS depressant effects of Meclizine can be potentiated by Ethanol or other CNS depressant agents
- Concurrent use of Benzodiazepines, Barbiturates, Tricyclic antidepressants, opiate agonists, skeletal muscle relaxants and antihistamines can potentiate the effects
- Concurrent use of other anticholinergics can potentiate the anticholinergic effects
- Meclizine can increase the absorption of digoxin by decreasing gastrointestinal motility
Contraindications
- Contraindicated in patients who are hypersensitive to Meclizine Hydrochloride and Pyridoxine Hydrochloride
Side Effects
- Drowsiness
- Dry mouth
- Blurred vision (rare)
Pregnancy & Lactation
- Pregnancy Category B
- Large-scale human studies have not demonstrated adverse fetal effects
- Suggested as the drug of first choice in treating nausea and vomiting during pregnancy
- Safety for use in nursing mothers not established
Precautions & Warnings
- May impair ability to perform hazardous activities requiring mental alertness or physical coordination
- Patients should avoid alcoholic beverages while taking Meclizine
- Use with caution in patients with asthma, glaucoma or prostate gland enlargement
Overdose Effects
- Symptoms include extreme excitability, seizures, drowsiness, and hallucinations
- Treatment involves appropriate supportive and symptomatic treatment, consider dialysis
