Indications

Pharmacology
- Meclizine is a piperazine-derivative antihistamine
- Has antiemetic properties
- Has anticholinergic properties
- Has antihistaminic properties
- Reduces the sensitivity of the labyrinthine apparatus
- May act through nerve pathways to the vomiting center
- May affect peripheral nerve pathways
- May affect other CNS centers
- Pyridoxine is vitamin B-6
Dosage & Administration
- 25-50 mg daily for adults and children 12 years of age & over for nausea and vomiting
- 25-50 mg for motion sickness, 1 hour prior to travel
- 50 mg for radiation sickness, 2-12 hours prior to treatment
- 25-100 mg daily for vertigo
- 25-50 mg for prevention of nausea and vomiting associated with emergency contraceptive pill (ECP)
Interaction
- CNS depressant effects potentiated by ethanol or other CNS depressant agents
- Concurrent use with benzodiazepines, barbiturates, tricyclic antidepressants, opiate agonists, skeletal muscle relaxants, and antihistamines can potentiate effects
- Concurrent use with other anticholinergics can potentiate anticholinergic effects
- Can increase absorption of digoxin by decreasing gastrointestinal motility
Contraindications
- Contraindicated in patients who are hypersensitive to the ingredients

- Drowsiness
- Dry mouth
- Blurred vision (rare)
Pregnancy & Lactation
- Pregnancy Category B
- No demonstrated adverse fetal effects in large-scale human studies
- Suggested as lowest risk of teratogenicity
- Drug of first choice in treating nausea and vomiting during pregnancy
Precautions & Warnings
- May impair ability to perform hazardous activities requiring mental alertness or physical coordination
- Patients should avoid alcoholic beverages
- Use with caution in patients with asthma, glaucoma, or prostate gland enlargement
Overdose Effects
- Symptoms: Extreme excitability, seizures, drowsiness, hallucinations
- Treatment: Appropriate supportive and symptomatic treatment, consider dialysis
