Indications
- Amelioration of allergic reactions to blood or plasma
- Adjunct to epinephrine in anaphylaxis
- Treatment of uncomplicated allergic conditions
- Sedation and relief of apprehension
- Active treatment of motion sickness
- Prevention and control of nausea and vomiting
- Adjunct to analgesics for postoperative pain control
- Preoperative, postoperative, and obstetric sedation

Pharmacology
Blocks postsynaptic mesolimbic dopaminergic receptors, exhibits α-adrenergic blocking effect, competes with histamine for the H1-receptor
Dosage & Administration
- Average adult dose is 25 mg for allergic conditions
- Nighttime sedation may be achieved with 25 to 50 mg for hospitalized adult patients
- Usual adult dose for nausea and vomiting is 12.5 to 25 mg
- Adjunct to preoperative or postoperative medication is 25 to 50 mg for adults
Interaction
- May intensify the sedative action of CNS depressants
- Epinephrine should not be used in case of Phenerex injection overdose
Contraindications
- Contraindicated in comatose states
- Should not be given by intra-arterial or subcutaneous route
Side Effects
- Drowsiness
- Extrapyramidal reactions
- Tachycardia, bradycardia
- Nausea, vomiting
- Allergic reactions like urticaria and dermatitis
- Leukopenia and agranulocytosis
- Thrombocytopenic purpura and jaundice
- Tissue necrosis with subcutaneous injection
Pregnancy & Lactation
Pregnancy Category C, should be used during pregnancy only if the potential benefit justifies the potential risk to the fetus
Precautions & Warnings
- Use with caution in patients with narrow-angle glaucoma
- No long term animal studies for carcinogenic potential, mutagenicity, or impairment of fertility
Use in Special Populations
- Contraindicated for children under 2
- Dosage should not exceed half the suggested adult dose in pediatric patients
- Dosage should be reduced for geriatric patients
Overdose Effects
- Range from mild depression to profound hypotension and unconsciousness
- Convulsions may rarely occur
- Treatment is essentially symptomatic and supportive
