Description
- Oral soluble film designed to be applied on top of the tongue
- Dissolves within 20 seconds and then is swallowed with saliva

Indications
- Prevention of nausea and vomiting associated with initial and repeat courses of emetogenic cancer chemotherapy
- Prevention and treatment of post-operative nausea and vomiting
- Prevention of radiotherapy-induced nausea and vomiting
Pharmacology
- Potent, highly selective 5HT3 receptor-antagonist
- Exact mode of action is not fully known
- Blocks initiation of vomiting reflex induced by chemotherapy and radiotherapy
Dosage
- Dosage varies for different indications and patient age/weight
- Available in different forms including oral tablets, orodispersible tablets, injection, and oral solution
Administration
- Orally dissolving film application steps: Tear pouch, place film on top of the tongue, allow to dissolve, swallow with or without liquid, wash hands after consumption
Interaction
- Metabolized by hepatic cytochrome P-450 drug-metabolizing enzymes
- No dosage adjustment recommended for patients on drugs metabolized by same enzymes
Contraindications
- Hypersensitivity to the drug or any of its components
- Concomitant use of apomorphine
Side Effects
- Frequently reported adverse events: headache, constipation, diarrhea
- Rare cases of anaphylaxis, brochospasm, tachycardia, hypokalemia
- No evidence of extrapyramidal reactions
Pregnancy & Lactation
- No evidence of carcinogenic, mutagenic, or fertility effects at recommended doses in animal studies
- No adequate and well-controlled studies in pregnant women
- Excreted in the breast milk of rats
Precautions & Warnings
- Hypersensitivity reactions reported in patients with hypersensitivity to other selective 5-HT3 receptor antagonists
- Not a drug that stimulates gastric or intestinal peristalsis
- Caution required in patients following abdominal surgery or with chemotherapy-induced nausea and vomiting
Use in Special Populations
- No dosage adjustment needed for patients with impaired renal function
- Specific dosage recommendations for patients with impaired hepatic function
- Limited information available about dosage in pediatric patients 4 years of age or younger
- No dosage adjustment needed for patients over the age of 65
