Indications
- Prevention of nausea and vomiting associated with emetogenic cancer chemotherapy
- Prevention and treatment of post-operative nausea and vomiting
- Prevention of radiotherapy-induced nausea and vomiting
Dosage
- Available in 4 mg and 8 mg orodispersible tablets
- Dosage based on patient weight and condition
- Also available in oral solution form for pediatric patients
Mode of Action
- Potent, highly selective 5HT3 receptor-antagonist
- Blocks initiation of vomiting reflex by activating vagal afferents via 5HT3 receptors
- Antagonizes 5HT3 receptors in both peripheral and central nervous system

Pharmacological Class

Administration
- Oral soluble film dissolves on top of the tongue within 20 seconds
- No water required for dissolution or swallowing
- Can be swallowed with or without liquid
Storage Conditions
- Store at temperature not exceeding 30°C
- Protect from light and moisture
Interaction
- Metabolized by hepatic cytochrome P-450 drug-metabolizing enzymes
- Clearance and half-life may be affected by inducers or inhibitors of these enzymes
Contraindications
- Hypersensitivity to the drug or any of its components
- Concomitant use of apomorphine
Side Effects
- Frequently reported adverse events include headache, constipation, and diarrhea
- Rash reported in approximately 1% of patients
- Rare cases of anaphylaxis, tachycardia, and hypokalemia
Pregnancy & Lactation
- Not mutagenic in standard tests for mutagenicity
- No evidence of impaired fertility or harm to the fetus in animal studies
- Caution advised when administered to nursing women
Precautions & Warnings
- Caution in patients with hypersensitivity to other 5-HT3 receptor antagonists
- Should not be used instead of nasogastric suction
- May mask a progressive ileus and/or gastric distension
Special Populations
- Dosage adjustment not needed for patients with impaired renal function
- Specific dosage recommendation for patients with severe hepatic impairment
