Indications
- Prevention of nausea and vomiting associated with initial and repeat courses of emetogenic cancer chemotherapy
- Prevention and treatment of post-operative nausea and vomiting
- Prevention of radiotherapy-induced nausea and vomiting
Description
- Orally dissolving film designed to be applied on top of the tongue
- Dissolves within 20 seconds
- Does not require water for dissolution or swallowing
- Active ingredient: Ondansetron base
- Molecular weight: 293.3
Pharmacology
- Serotonin subtype 3 (5HT3) receptor-antagonist
- Blocks 5HT3 receptors in the peripheral and central nervous system
- Mechanisms of action in post-operative nausea and vomiting are not known
Dosage
- Various dosages for adults, pediatric patients, adults/gertiatric/child of 12 years or over, pediatric (4-11 years)
- Dosage adjustment for different chemotherapeutic agents and radiotherapy
Administration
- Tear the pouch carefully
- Place the film on top of the tongue
- Do not chew or swallow the film whole
- Swallow after the film dissolves
- Wash hands after taking the film
Interaction
- Osetron does not induce or inhibit the cytochrome P-450 drug-metabolizing enzyme system of the liver
- Metabolized by hepatic cytochrome P-450 drug-metabolizing enzymes
Contraindications
- Hypersensitivity to the drug or any of its components
- Concomitant use of apomorphine
Side Effects
- Frequently reported adverse events: headache, constipation, diarrhea, rash
- Other reported adverse events: sensation of flushing or warmth, hiccups, liver enzyme abnormalities, anaphylaxis, brochospasm, tachycardia, angina, hypokalemia, shortness of breath
- No evidence of extrapyramidal reactions
Pregnancy & Lactation
- No evidence of carcinogenic effects in rats and mice
- Not mutagenic in standard tests
- No impairment of fertility or harm to the fetus in rats and rabbits
- Caution advised for nursing women
Precautions & Warnings
- Hypersensitivity reactions reported in patients hypersensitive to other selective 5-HT3 receptor antagonists
- Not a drug that stimulates gastric or intestinal peristalsis
- Caution in patients following abdominal surgery or with chemotherapy-induced nausea and vomiting
Use in Special Populations
- Dosage adjustment for patients with impaired renal function
- Dosage adjustment for patients with impaired hepatic function
- Limited information for pediatric patients 4 years of age or younger
