Indications
- Prevention of nausea and vomiting associated with initial and repeat courses of emetogenic cancer chemotherapy
- Prevention and treatment of post-operative nausea and vomiting
- Prevention of radiotherapy-induced nausea and vomiting
Dosage
- Different dosage forms for adults, pediatric patients, and cancer chemotherapy
- Dosage adjustment for patients with impaired renal function
- Dosage adjustment for patients with impaired hepatic function
- Limited information about dosage for pediatric patients 4 years of age or younger
- No dosage adjustment needed in patients over the age of 65
Administration
- Orally dissolving film designed to be applied on top of the tongue and dissolve within 20 seconds
- Does not require water to aid dissolution or swallowing
- Specific steps for administration including tearing the pouch, placing film on the tongue, and swallowing after dissolution
Storage Conditions
- Store at a temperature not exceeding 30°C in a dry place

Pharmacology
- Potent, highly selective 5HT3 receptor-antagonist
- Precise mode of action in the control of nausea and vomiting is not fully known
- Action likely due to antagonism of 5HT3 receptors on neurons in the peripheral and central nervous system
Interaction
- Does not induce or inhibit the cytochrome P-450 drug-metabolizing enzyme system of the liver
- Metabolized by hepatic cytochrome P-450 drug-metabolizing enzymes
- Inducers or inhibitors of these enzymes may change the clearance and half-life of the medication
Contraindications
- Contraindicated in patients known to have hypersensitivity to the drug or any of its components
- Concomitant use of apomorphine
Side Effects
- Frequently reported adverse events including headache, constipation, and diarrhea
- Rare cases of anaphylaxis, bronchospasm, tachycardia, angina, and hypokalemia reported
- No evidence of extrapyramidal reactions, except for rare cases of oculogyric crisis
Pregnancy & Lactation
- No evidence of impaired fertility or harm to the fetus in animal studies
- No adequate and well-controlled studies in pregnant women
- Excreted in the breast milk of rats
Precautions & Warnings
- Hypersensitivity reactions reported in patients who have exhibited hypersensitivity to other selective 5-HT3 receptor antagonists
- Not a drug that stimulates gastric or intestinal peristalsis
- Caution needed in patients following abdominal surgery or with chemotherapy-induced nausea and vomiting
Use in Special Populations
- Dosage recommendation for patients with impaired renal function is the same as for the general population
- Dosage adjustment for patients with severe hepatic impairment
- Limited information about dosage in pediatric patients 4 years of age or younger
