Indications

Description
- Orally dissolving film designed to be applied on top of the tongue
- Dissolves within 20 seconds
- Active ingredient is Zofra ODT base, a selective blocking agent of the serotonin 5-HT3 receptor type
- Empirical formula is C18H19N3O representing a molecular weight of 293.3
Pharmacology
- Potent, highly selective 5HT3 receptor-antagonist
- Exact mode of action in control of nausea and vomiting is not known
- Blocks initiation of vomiting reflex by activating vagal afferents via 5HT3 receptors

- Adult dosage for chemotherapy-induced Nausea and Vomiting

Interaction
- Zofra ODT does not itself appear to induce or inhibit the cytochrome P-450 drug-metabolizing enzyme system of the liver
- Metabolized by hepatic cytochrome P-450 drug-metabolizing enzymes
Contraindications
- Contraindicated in patients known to have hypersensitivity to the drug or any of its components
- Concomitant use of apomorphine
Side Effects
- Frequently reported adverse events
- Occurrence of rash, sensation of flushing or warmth, hiccups, liver enzyme abnormalities
- Rare cases of anaphylaxis, bronchospasm, tachycardia, angina, hypokalemia, shortness of breath
Pregnancy & Lactation
- No evidence of impaired fertility or harm to the fetus in rats and rabbits at certain oral doses
- No adequate and well-controlled studies in pregnant women
- Excreted in the breast milk of rats
- Caution should be exercised when administered to nursing women
Precautions & Warnings
- Hypersensitivity reactions reported in patients with hypersensitivity to other selective 5-HT3 receptor antagonists
- Should not be used instead of nasogastric suction
- May mask a progressive ileus and/or gastric distension
Use in Special Populations
- Dosage adjustment for patients with impaired renal function
- Dosage adjustment for patients with impaired hepatic function
- Little information about dosage in pediatric patients 4 years of age or younger
