  • Acute and delayed nausea and vomiting
  • Uncontrolled nausea and vomiting
  • Chemotherapy-induced nausea and vomiting (CINV)
  • Radiotherapy-induced nausea and vomiting (RINV)
  • Post-operative & Post-discharge nausea and vomiting (PONV & PDNV)

Mode of Action

  • 5-HT3 receptor antagonist
  • Strong binding affinity for 5-HT3 receptor
  • Little or no affinity for other receptors
  • Blocks the actions of serotonin at 5-HT3 receptor
  • Influence on postoperative nausea and vomiting

Pharmacokinetics

  • Exhibits linear dose-proportional pharmacokinetics
  • Metabolized via kidney and liver
  • Longer half-life of 40 hours
  • Greater 5-HT3 receptor binding affinity

Dosage and Administration

  • Usual adult tablet dosage of 0.5 mg daily
  • Single IV dose of 0.075 mg over 10 seconds for adults
  • Different dosages for chemotherapy-induced nausea and vomiting, radiotherapy-induced nausea and vomiting, post-operative nausea and vomiting, and children dosage based on body weight

Interactions

  • Safe administration with corticosteroids, analgesics, antiemetics/antinauseants, antispasmodics, and anticholinergic agents
  • No significant pharmacokinetic interactions with metoclopramide
  • Low potential for clinically significant drug interactions

Contraindications

  • Contraindicated in patients with hypersensitivity to the drug or any of its components

Side Effects

  • Most common adverse reactions are headaches and constipation

Pregnancy and Lactation

  • Pregnancy category B
  • Not known whether excreted in breast milk

Use in Special Populations

  • No dosage adjustment recommended in elderly patients >65 years of age
  • Specific dosage for use in children based on age and body weight
  • No dosage adjustment recommended in patients with renal and hepatic dysfunction

Overdose Effects

  • No known antidote to Paloron
  • Overdose should be managed with supportive care
