Indications
- Acute and delayed nausea and vomiting
- Uncontrolled nausea and vomiting
- Chemotherapy-induced nausea and vomiting (CINV)
- Radiotherapy-induced nausea and vomiting (RINV)
- Post-operative & Post-discharge nausea and vomiting (PONV & PDNV)
Pharmacology
- 5-HT3 receptor antagonist
- Strong binding affinity for 5-HT3 receptors
- Little or no affinity for other receptors
- Works by blocking the actions of Serotonin
- Influences postoperative nausea and vomiting
- Exhibits linear dose-proportional pharmacokinetics
Dosage & Administration
- Usual adult tablet dosage of 0.5 mg daily
- Usual adult IV dosage of 0.075 mg over 10 seconds
- Specific dosages for chemotherapy-induced nausea and vomiting
- Specific dosage for radiotherapy-induced nausea and vomiting
- Specific dosage for post-operative nausea and vomiting
- Children dosage based on body weight
Interaction
- Safely administered with corticosteroids, analgesics, antiemetics/antinauseants, antispasmodics and anticholinergic agents
- No significant pharmacokinetic interactions with metoclopramide
- Not inhibitor of various CYP enzymes
- Low potential for clinically significant drug interactions
Contraindications
- Contraindicated in patients with hypersensitivity to the drug or any of its components
Side Effects
- Common adverse reactions include headaches and constipation
Pregnancy & Lactation
- Pregnancy category 'B'
- Excretion in breast milk not known
Use in Special Populations
- No dosage adjustment recommended in elderly patients >65 years of age
- Specific dosage for children (1 month to 10 years)
- No dosage adjustment recommended in patients with renal and hepatic dysfunction
Overdose Effects
- No known antidote
- Manage with supportive care
