Description

  • Palostat is indicated for acute and delayed nausea and vomiting, uncontrolled nausea and vomiting, chemotherapy-induced nausea and vomiting, radiotherapy-induced nausea and vomiting, post-operative and post-discharge nausea and vomiting. It works as a 5-HT3 receptor antagonist.

DosageAndAdministration

  • Usual dosage: Adult tablet dosage - 0.5 mg daily, Adult IV dosage - 0.075 mg over 10 seconds. Children dosage: A single IV dose at 20 mcg/kg body weight with maximum dose of 1.5 mg.

Pharmacology

  • Palonosetron is a 5-HT3 receptor antagonist with a strong binding affinity for this receptor. It binds to the 5-HT3 receptor and blocks the actions of serotonin, associated with nausea and vomiting.

Interaction

  • Palostat has been safely administered with corticosteroids, analgesics, antiemetics/antinauseants, antispasmodics, and anticholinergic agents. Concomitant administration of Palostat and metoclopramide has no significant pharmacokinetic interactions.

Contraindications

  • Palostat is contraindicated in patients known to have hypersensitivity to the drug or any of its components.

SideEffects

  • The most common adverse reactions are headaches and constipation.

PregnancyAndLactation

  • Pregnancy category 'B'. It is not known whether Palonosetron is excreted in breast milk.

SpecialPopulations

  • No dosage adjustment is recommended in elderly patients over 65 years of age. No dosage adjustment is recommended in patients with renal and hepatic dysfunction.

OverdoseEffects

  • There is no known antidote to Palostat. Overdose should be managed with supportive care.

  • Store in a cool and dry place, protected from light.
