Description
Paloxiron is indicated in the treatment and prevention of acute and delayed nausea and vomiting caused by chemotherapy, radiotherapy, post-operative and post-discharge conditions.

- Mode of Action: Acts as a 5-HT3 receptor antagonist by blocking the actions of Serotonin associated with nausea and vomiting.
- Pharmacology:
- Exhibits linear dose-proportional pharmacokinetics
- Has a longer half-life (40 hours)
- Metabolized via kidney and liver CYP2D6, CYP3A4, and CYP1A2 isoenzymes
- Dosage & Administration:
- Usual adult tablet dosage: 0.5 mg daily
- Usual adult IV dosage: A single IV dose of 0.075 mg administered over 10 seconds
- Variations in dosage for different conditions and age groups
- Interaction: Safely administered with corticosteroids, analgesics, antiemetics, and other medications based on clinical trials. Low potential for clinically significant drug interactions.
- Contraindications: Contraindicated in patients known to have hypersensitivity to the drug or any of its components.
- Side Effects:
- Common adverse reactions include headaches and constipation
- Pregnancy & Lactation: Pregnancy category 'B'. Excretion in breast milk is not known.
- Use in Special Populations:
- No dosage adjustment recommended in elderly patients >65 years of age
- Specific dosage instructions for children
- No dosage adjustment recommended in patients with renal and hepatic dysfunction
- Overdose Effects: No known antidote to Paloxiron. Overdose should be managed with supportive care.
