Description
- Palzen is indicated for the management of acute and delayed nausea and vomiting, including uncontrolled nausea and vomiting, chemotherapy-induced nausea and vomiting (CINV), radiotherapy-induced nausea and vomiting (RINV), post-operative and post-discharge nausea and vomiting (PONV & PDNV). It is a 5-HT3 receptor antagonist with a strong binding affinity for this receptor and little or no affinity for other receptors. Palonosetron exhibits linear dose-proportional pharmacokinetics over the dose range 1-90 μg/kg in healthy subjects and in patients with cancer. It has a longer half-life (40 hours) and a greater 5-HT3 receptor binding affinity. Usual adult tablet dosage is 0.5 mg daily, while the usual adult IV dosage is a single IV dose of 0.075 mg administered over 10 seconds. It is safe to administer with corticosteroids, analgesics, and other antiemetics/antinauseants. Palzen is contraindicated in patients known to have hypersensitivity to the drug or any of its components. The most common adverse reactions are headaches and constipation. It is classified as pregnancy category 'B' and its excretion in breast milk is not known. No dosage adjustment is recommended in elderly patients >65 years of age. In case of overdose, there is no known antidote to Palzen and it should be managed with supportive care. Palzen belongs to the therapeutic class of anti-emetic drugs and should be stored in a cool and dry place, protected from light.
