Indications

  • Partial and secondary generalized tonic-clonic seizures
  • Primary generalized tonic-clonic seizures
  • Trigeminal neuralgia
  • Prophylaxis of bipolar disorder

Pharmacology

  • Depresses activity in the nucleus ventralis of the thalamus
  • Reduces synaptic propagation of excitatory impulses
  • Decreases summation of temporal stimulation
  • Lowers neural discharge by limiting influx of Na ions across cell membrane
  • Stimulates the release of antidiuretic hormone (ADH)
  • Potentiates the action of ADH in promoting reabsorption of water

Dosage & Administration

  • Initial dosage for adults and children over 12 years of age
  • Increase dosage at weekly intervals
  • Combination therapy with other anticonvulsants
  • Dosage for trigeminal neuralgia
  • Maintenance dosage for various age groups

Interaction

  • Potential interaction with oral contraceptives
  • Interaction with hepatic enzyme inducers like Phenytoin
  • Possible increase in dosage requirement

Contraindications

  • Allergy to any of the ingredients
  • Atrioventricular block
  • History of bone marrow depression
  • Hereditary blood disorders (porphyrias)
