Description
- Indicated for the treatment of panic disorder with or without agoraphobia
- Also used in the treatment of Lennox-Gastaut Syndrome, akinetic and myoclonic seizures
- Exhibits anticonvulsive, sedative, muscle relaxing, and anxiolytic effects
- Enhances GABAergic neurotransmission at inhibitory synapses
Dosage & Administration
- Initial adult dose for seizure disorders should not exceed 1.5 mg/day divided into three doses
- Initial adult dose for panic disorder is 0.25 mg given in two divided doses
- Pediatric patients initial dose should be between 0.01 and 0.03 mg/kg/day
Interaction
- Does not alter the pharmacokinetics of phenytoin, carbamazepine, or phenobarbital
Contraindications
- Not to be used in patients with hypersensitivity to benzodiazepines
- Contraindicated in acute narrow angle glaucoma
Side Effects
- Frequently occurring side effects include CNS depression, drowsiness, and ataxia
- Behavior problems, abnormal eye movements, tremor, vertigo, confusion, depression, and insomnia may occur
Pregnancy & Lactation
- Use during pregnancy may cause congenital malformations
- Act cautiously during breastfeeding
Precautions & Warnings
- May increase the incidence of generalized tonic-clonic seizures in certain patients
- Concomitant use with valproic acid may produce absence status
Use in Special Populations
- Caution in pediatric use due to increased production of saliva and bronchial secretion
- Benzodiazepine effects may be greater in elderly patients
- Renal impairment does not affect the pharmacokinetics
- Plasma protein binding in cirrhotic patients is significantly different
Overdose Effects
- May cause areflexia, apnoea, hypotension, and cardiorespiratory depression
- Common symptoms include difficulty in sleeping, changes in mood, and increased sweating

Reconstitution
- Must be diluted with water for injection prior to intravenous administration
- IV injection should be administered slowly with continuous monitoring
- Infusion bags other than PVC should be used for infusing
