Indications
- Neuropathic Pain
- Adjunctive therapy in partial seizure and secondary generalized seizure
Pharmacology
- Anti-convulsant
- Structural analog of gamma-amino-butyric-acid (GABA)
- Binds with alpha-2-delta subunit of voltage-gated L-type Calcium channel
- Inhibits branched chain amino acid transferase
- Inhibits neurotransmitter release of excitatory amino acid
Dosage
- Initiate with 300 mg dose on Day-1
- Titrate up to 1800 mg daily dose for Neuropathic Pain
- Effective dose for patients over 12 years: 900 to 1800 mg/day
- Starting dose for paediatric patients age 3-12 years: 10-15 mg/kg/day
- Divided doses and upward titration
Administration
- Oral administration
- Can be taken with or without food
Interaction
- Antacids reduce bioavailability by up to 20%
- Cimetidine may alter reanal excretion
- No interaction with other anti-epileptic drugs or oral contraceptive preparations
Contraindications
- Known hypersensitivity to the drug
