Indications
- Neuropathic Pain
- Adjunctive therapy in partial seizure and secondary generalized seizure
Pharmacology
- Structural analog of gamma-amino-butyric-acid (GABA)
- Binds with the alpha-2-delta subunit of voltage gated L-type Calcium channel
- Inhibits branched chain amino acid transferase
- Inhibits neurotransmitter release of excitatory amino acid
Dosage
- 300 mg dose on Day-1
- 300 mg twice on Day-2
- 300 mg thrice on Day-3
- Daily dose of 1800 mg
- 900 to 1800 mg/day for patients over 12 years of age
- Starting dose of 300 mg three times a day for patients over 12 years of age
- Starting dose ranging from 10-15 mg/kg/day for pediatric patients age 3-12 years
Administration
Oral administration with or without food
Interaction
- Reduction of bioavailability by up to 20% with antacids
- Alteration of renal excretion by cimetidine
- No interaction with other anti-epileptic drugs or oral contraceptive preparations
