Indications
- Neuropathic Pain
- Adjunctive therapy in partial seizure and secondary generalized seizure
Pharmacology
- Anti-convulsant
- Structural analog of gamma-amino-butyric-acid (GABA)
- Binds with the alpha-2-delta subunit of voltage gated L-type Calcium channel
- Inhibits branched chain amino acid transferase
- Inhibits neurotransmitter release of excitatory amino acid
Dosage
- Initiation: single 300 mg dose on Day-1, 300 mg twice on Day-2, 300 mg thrice on Day-3
- Subsequent titration up to a daily dose of 1800 mg (divided TID)
- Epilepsy: Effective dose of 900 to 1800 mg/day given in three divided doses
- Starting dose for pediatric patients: 10-15 mg/kg/day in 3 divided doses
Administration
Oral administration with or without food
Interaction
- Antacids may reduce bioavailability by up to 20%
- Cimetidine may alter renal excretion
- No interaction with other anti-epileptic drugs or oral contraceptive preparations
