Description

  • Phenyltriazine antiepileptic medication
  • Used to control epileptic seizures
  • Also used to treat some types of bipolar disorder
  • Inhibits voltage-sensitive sodium channels
  • Stabilizes neuronal membranes
  • Inhibits presynaptic excitatory neurotransmitter release

Pharmacology

  • Inhibition of voltage-sensitive sodium channels
  • Modulation of release of presynaptic excitatory neurotransmitters
  • Binding properties to various receptors
  • Weak inhibitory effects on serotonin 5-HT3 receptors
  • Weak binding to adenosine, adrenergic, dopamine, GABA, histamine, opioid, mACh, and serotonin 5-HT2 receptors
  • Inhibition of Cav2.3 (R-type) calcium currents

  • Different escalation regimens for patients with epilepsy aged 2 years and older

Interaction

  • Increased concentrations with valproate
  • Decreased concentrations with carbamazepine, phenytoin, phenobarbital, primidone, and rifampin
  • Decreased concentrations with estrogen-containing oral contraceptives
  • Decreased exposure with protease inhibitors lopinavir/ritonavir and atazanavir/ritonavir

Overdose Effects
