Indications

Description
- Phenyltriazine antiepileptic
- Treats some types of epilepsy
- Controls epileptic seizures
- Inhibits voltage-sensitive sodium channels
- Stabilizes neuronal membranes
- Inhibits presynaptic excitatory neurotransmitter release
Pharmacology
- Mechanism of action not fully elucidated
- Chemically unrelated to phenytoin and carbamazepine
- Resembles actions of phenytoin and carbamazepine
- Inhibition of sodium channels
- Suppression of glutamate release
- Binding properties to various receptors
- Weak effects on serotonin, adrenergic, dopamine, GABA, histamine, opioid, mACh, and sigma receptors
- Clinical benefit in neuropathic pain states

Interaction
- Valproate increases Lamotil concentrations by more than 2-fold
- Carbamazepine, phenytoin, phenobarbital, primidone, and rifampin decrease Lamotil concentrations by approximately 40%
- Estrogen-containing oral contraceptives decrease Lamotil concentrations by approximately 50%
- Protease inhibitors lopinavir/ritonavir and atazanavir/ritonavir decrease Lamotil exposure

Use in Special Populations
- Safety and effectiveness not demonstrated in very young pediatric patients aged 1 to 24 months for epilepsy
- Safety and effectiveness not established in pediatric patients aged 10 to 17 years for bipolar disorder
Overdose Effects
