Indications

  • Monotherapy for partial-onset seizures in adults and adolescents
  • Adjunctive therapy for partial-onset seizures in adults, adolescents, children, and infants

Description

  • Precise mechanism(s) unknown
  • Minimal activity in submaximal stimulation and threshold tests
  • Protection against secondarily generalized activity from focal seizures induced by pilocarpine and kainic acid
  • Inhibition of burst firing without affecting normal neuronal excitability

Dosage

  • Recommended starting dose for adults and adolescents from 16 years of age
  • Initial therapeutic dose for adults and adolescents (≥18 years) and adolescents (12 to 17 years) weighing 50 kg or more
  • Pediatric population dosage and monotherapy/adjuvant therapy details
  • Administration in two equally divided doses

Administration

  • Oral administration
  • Swallowed with sufficient quantity of liquid
  • May be taken with or without food

Interaction

  • No influence on serum concentrations of existing antiepileptic medicinal products
  • Interaction with Probenecid, Methotrexate, Laxatives, Food, and Alcohol

Contraindications

  • Hypersensitivity to the active substance or other pyrrolidone derivatives or to any of the excipients

Side Effects

  • Most common adverse reactions in adult and pediatric patients

Pregnancy & Lactation

  • Not recommended during pregnancy and in women of childbearing potential not using contraception unless clinically necessary
  • Excretion in human breast milk
  • Breastfeeding not recommended

Precautions & Warnings

  • Dose adjustment in patients with renal impairment
  • Rare cases of decreased blood cell counts
  • Association with acute kidney injury
  • Monitoring for signs of depression and/or suicidal ideation and behaviors

Use in Special Populations

  • Elderly patients with compromised renal function
  • Individualization of daily dose according to renal function in elderly patients, patients with renal impairment, and hepatic impairment

Overdose Effects

  • Observation of somnolence, agitation, aggression, depressed level of consciousness, respiratory depression, and coma
  • No specific antidote
  • Induction of emesis
  • Hemodialysis
