Indications

  • Monotherapy in the treatment of partial seizures in adults
  • Adjunctive therapy in the treatment of partial seizures in adults
  • Monotherapy in the treatment of partial seizures in children aged 4-16 years
  • Adjunctive therapy in the treatment of partial seizures in children aged 2-16 years

Pharmacology

  • Primary activity exerted through the metabolite derivative (MHD) of Oxcarbazepine
  • Mechanism of action based on blockade of voltage-sensitive sodium channels
  • Stabilization of hyper excited neural membranes
  • Inhibition of repetitive neuronal firing
  • Diminishment of propagation of synaptic impulses

Dosage & Administration

  • Recommended daily dose for adults is 1200 mg/day
  • Maximum daily dose for adults is 2400 mg/day
  • Dosage initiation with 600 mg/day for adults
  • Dosage initiation with 8 to 10 mg/kg/day for pediatrics aged 2 and above
  • Dosage initiation with 16 to 20 mg/kg/day for pediatrics aged 4 and below
  • Dosage adjustment for patients with creatinine clearance <30 mL/min
  • Conversion to monotherapy with gradual withdrawal of concomitant antiepileptic drugs

Interaction

  • Inhibition of the enzyme CYP2C19 by Trileptal and its metabolite
  • Potential interactions with medicinal products metabolized by CYP2C19
  • Reduced effectiveness of hormonal contraceptives when co-administered with Trileptal

Contraindications

  • Hypersensitivity to the active substance or any of the excipients

Side Effects

  • Common adverse reactions: somnolence, headache, dizziness, diplopia, nausea, vomiting, fatigue
  • Rare occurrence of clinically significant hyponatremia
  • Reports of Class I (immediate) hypersensitivity reactions

Pregnancy & Lactation

  • Potential for serious birth defects (e.g., cleft palate) when administered during pregnancy
  • Transmission of Oxcarbazepine and its active metabolite (MHD) to the newborn
  • Excretion of Oxcarbazepine and MHD in human breast milk

Precautions & Warnings

  • Caution advised when combining alcohol with Oxazep therapy due to possible additive sedative effect
  • Gradual withdrawal recommended to minimize potential for increased seizure frequency
