Indications

Description
- Barbiturate
- Nonselective central nervous system depressant
- Primarily used as a sedative hypnotic
- Also used as an anticonvulsant in subhypnotic doses
Pharmacology
- Acts on GABA receptors
- Increases synaptic inhibition
- Elevates seizure threshold
- Reduces spread of seizure activity
- Inhibits calcium channels
- Decreases excitatory transmitter release
- Likely affects the polysynaptic midbrain reticular formation
Dosage & Administration
- Suggested pediatric and adult oral doses
- Suggested pediatric and adult injection doses
- Specific doses for preoperative, anticonvulsant, daytime sedative, bedtime hypnotic, preoperative sedation, insomnia, convulsions, status epilepticus
Interaction
- Additive CNS depressant effects with alcohol or other CNS depressants
Contraindications
- Patients with known sensitivity to Phenobarbital
- Patients with a history of latent porphyria
Side Effects
- Most common adverse reaction: somnolence
- Less frequent adverse reactions: agitation, confusion, hyperkinesia, ataxia, CNS depression, nightmares, nervousness, psychiatric disturbance, hallucinations, insomnia, anxiety, dizziness, thinking abnormality, apnea, bradycardia, hypotension, nausea, vomiting, constipation

Precautions & Warnings
- Tolerance and dependence may occur with continuing use
- Caution with mentally depressed patients
- Caution with patients with suicidal tendencies or a history of drug abuse

- Manifested by CNS and respiratory depression
- Supportive treatment for overdose
