Indications
- Neuropathic pain associated with diabetic peripheral neuropathy (DPN)
- Postherpetic neuralgia (PHN)
- Adjunctive therapy for the treatment of partial-onset seizures in patients 1 month of age and older
- Fibromyalgia
- Neuropathic pain associated with spinal cord injury
Pharmacology
- Structural derivative of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA)
- Binds with high affinity to the alpha2-delta site in central nervous system tissues
Dosage
- Maximum recommended dose of Pregabalin is 100 mg three times a day
- Dosing should begin at 50 mg three times a day
- Dose may be increased based on efficacy and tolerability
- Recommended dose of Pregabalin for postherpetic neuralgia is 75 to 150 mg two times a day or 50 to 100 mg three times a day
- Dosage should be adjusted based on individual patient response and tolerability
Administration
- Oral route of administration
- Can be taken with or without food
- Pregabalin CR tablet should be administered after an evening meal
- Should be swallowed whole and not split, crushed, or chewed
Interaction
- Unlikely to be involved in significant pharmacokinetic drug interactions
- No drug interactions with food and others
Contraindications
- Contraindicated in patients with known hypersensitivity to Pregabalin or any of its components
Side Effects
- Common side effects include dizziness, somnolence, dry mouth, edema, blurred vision, weight gain
- In pediatric patients, side effects may include increased weight and increased appetite
Pregnancy & Lactation
- No adequate and well-controlled studies in pregnant women
- Small amounts detected in the milk of lactating women
- Breastfeeding not recommended during treatment
Precautions & Warnings
- Angioedema can occur and may be associated with life threatening respiratory compromise
- Discontinue immediately if hypersensitivity reactions occur
- Increased risk of suicidal thoughts or behavior
- Respiratory depression may occur with concomitant CNS depressants
- May cause dizziness and somnolence, impairing ability to drive or operate machinery
Use in Special Populations
- Safety and effectiveness not established in pediatric patients for certain indications
- Safety and effectiveness in pediatric patients below the age of 1 month not established
Overdose Effects
- Signs and symptoms may include reduced consciousness, depression/anxiety, confusional state, agitation, seizures, heart block
