Medicine Details

Indications

  • Neuropathic pain associated with diabetic peripheral neuropathy (DPN)
  • Postherpetic neuralgia (PHN)
  • Adjunctive therapy for the treatment of partial-onset seizures in patients 1 month of age and older
  • Fibromyalgia
  • Neuropathic pain associated with spinal cord injury

Pharmacology

  • Structural derivative of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA)
  • Binds with high affinity to the alpha2-delta site in central nervous system tissues

Dosage

  • Maximum recommended dose of Pregabalin is 100 mg three times a day
  • Dosing should begin at 50 mg three times a day
  • Dose may be increased based on efficacy and tolerability
  • Recommended dose of Pregabalin for postherpetic neuralgia is 75 to 150 mg two times a day or 50 to 100 mg three times a day
  • Dosage should be adjusted based on individual patient response and tolerability

Administration

  • Oral route of administration
  • Can be taken with or without food
  • Pregabalin CR tablet should be administered after an evening meal
  • Should be swallowed whole and not split, crushed, or chewed

Interaction

  • Unlikely to be involved in significant pharmacokinetic drug interactions

Contraindications

  • Contraindicated in patients with known hypersensitivity to Pregabalin or any of its components

Side Effects

  • Common side effects include dizziness, somnolence, dry mouth, edema, blurred vision, weight gain

Pregnancy & Lactation

  • No adequate and well-controlled studies in pregnant women
  • Small amounts detected in the milk of lactating women
  • Breastfeeding not recommended during treatment

Precautions & Warnings

  • Angioedema can occur and may be associated with life threatening respiratory compromise
  • Discontinue immediately if hypersensitivity reactions occur
  • Increased risk of suicidal thoughts or behavior
  • Respiratory depression may occur with concomitant CNS depressants
  • May cause dizziness and somnolence, impairing ability to drive or operate machinery

Use in Special Populations

  • Safety and effectiveness not established in pediatric patients for certain indications
  • Safety and effectiveness in pediatric patients below the age of 1 month not established

Overdose Effects

  • Signs and symptoms may include reduced consciousness, depression/anxiety, confusional state, agitation, seizures, heart block
