Indications
- Neuropathic pain associated with diabetic peripheral neuropathy (DPN)
- Postherpetic neuralgia (PHN)
- Adjunctive therapy for the treatment of partial-onset seizures in patients 1 month of age and older
- Fibromyalgia
- Neuropathic pain associated with spinal cord injury
Pharmacology
- Mechanism of Action:
- Structural derivative of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA)
- Binds with high affinity to the alpha2-delta site in central nervous system tissues
- Antinociceptive and antiseizure effects
Dosage
- Neuropathic Pain (DPN) in Adults:
- Maximum recommended dose of Pregabalin: 100 mg three times a day
- Dosing should begin at 50 mg three times a day
- Postherpetic Neuralgia (PHN) in Adults:
- Recommended dose of Pregabalin: 75 to 150 mg two times a day
- Dosing should begin at 75 mg two times a day
- Fibromyalgia in Adults: Recommended dose of Pregabalin: 300 to 450 mg/day
- Neuropathic Pain associated with Spinal Cord Injury in Adults: Recommended dose range of Pregabalin: 150 to 600 mg/day
- Conversion from Pregabalin capsules to Pregabalin CR capsule tablet: Guidelines provided
Administration
- Route of Administration: Oral, with or without food
- Pregabalin CR Tablet Administration: After an evening meal, swallowed whole
- Discontinuation: Gradually tapered over a minimum of 1 week

- Medication: Unlikely to have significant pharmacokinetic interactions
- Food and Others: Not applicable

- Dry mouth
- Edema

- Increased weight
- Increased appetite
Pregnancy & Lactation
- Pregnant Women:
- No adequate and well-controlled studies
- Potential risk to fetus
- Lactating Women: Small amounts detected in milk, breastfeeding not recommended
Precautions & Warnings
- Angioedema
- Hypersensitivity reactions
- Risk of suicidal thoughts or behavior
- Respiratory depression
- Impairment of driving or operating machinery
- Increased seizure frequency on rapid discontinuation
- Peripheral edema
- Caution with coadministering with thiazolidinedione antidiabetic agents
