Indications
- Neuropathic pain associated with diabetic peripheral neuropathy (DPN)
- Postherpetic neuralgia (PHN)
- Adjunctive therapy for the treatment of partial-onset seizures in patients 1 month of age and older

Pharmacology
Pregabalin is a structural derivative of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA). It binds with high affinity to the alpha2-delta site in central nervous system tissues.
Dosage
- Maximum recommended dose: 100 mg three times a day for neuropathic pain associated with diabetic peripheral neuropathy
- Recommended dose for postherpetic neuralgia: 75 to 150 mg two times a day
- Recommended dose for fibromyalgia: 300 to 450 mg/day
- Recommended dose range for neuropathic pain associated with spinal cord injury: 150 to 600 mg/day
Administration
- Route of administration: Oral
- Special instruction for Pregabalin CR tablet administration: Swallowed whole after an evening meal
Interaction
- Drug interaction with medication: Unlikely to be involved in significant pharmacokinetic drug interactions
Contraindications
Contraindicated in patients with known hypersensitivity to Pregabalin or any of its components
Side Effects
- Dizziness
- Somnolence
- Dry mouth
- Edema
- Blurred vision
- Weight gain
- Difficulty with concentration/attention (thinking abnormal)
- Increased weight and increased appetite in pediatric patients
Pregnancy & Lactation
- Pregnancy: No adequate and well-controlled studies, potential risk to fetus
- Lactation: Small amounts detected in the milk, not recommended during treatment
Precautions & Warnings
- Angioedema and hypersensitivity reactions may occur
- Increased risk of suicidal thoughts or behavior
- Respiratory depression may occur when used with concomitant CNS depressants
- May cause dizziness and somnolence, impairing ability to drive or operate machinery
- Increased seizure frequency or other adverse reactions upon rapid discontinuation
- Possible peripheral edema
- Caution when coadministering with thiazolidinedione antidiabetic agents
Use in Special Populations
- Use in children and adolescents: Safety and effectiveness not established in various conditions
Overdose Effects
Signs and symptoms include reduced consciousness, depression/anxiety, seizures, and heart block. No specific antidote.
