Indications
- Neuropathic pain associated with diabetic peripheral neuropathy (DPN)
- Postherpetic neuralgia (PHN)
- Adjunctive therapy for the treatment of partial-onset seizures in patients 1 month of age and older
- Fibromyalgia
- Neuropathic pain associated with spinal cord injury
Pharmacology
Structural derivative of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA), binds with high affinity to the alpha2-delta site, involved in anti-nociceptive and antiseizure effects
Dosage
- Maximum recommended dose of 100 mg three times a day
- Recommended dose of 75 to 150 mg two times a day or 50 to 100 mg three times a day for postherpetic neuralgia
- Recommended dose of 300 to 450 mg/day for fibromyalgia
- Recommended dose range of 150 to 600 mg/day for neuropathic pain associated with spinal cord injury
Administration
- Route of administration: Oral, can be taken with or without food
- Pregabalin CR tablet administration: After an evening meal, swallowed whole and should not be split, crushed or chewed
- Discontinuation: Gradually tapered over a minimum of 1 week

- Medication interaction: Unlikely to be involved in significant pharmacokinetic drug interactions
- Food and others interaction: Not applicable
Contraindications
Contraindicated in patients with known hypersensitivity to Pregabalin
Side Effects
- Dizziness
- Somnolence
- Dry mouth
- Edema
- Blurred vision
- Weight gain
- Thinking abnormal (primarily difficulty with concentration/attention)
Pregnancy & Lactation
- Pregnancy: No adequate and well-controlled studies, potential risk to fetus
- Lactation: Small amounts detected in the milk of lactating women, not recommended during treatment
Precautions & Warnings
- Angioedema: Can occur and may be associated with life threatening respiratory compromise requiring emergency treatment
- Suicidal thoughts or behavior: Increased risk with Pregabalin, monitor and adjust dosage as appropriate
- Respiratory depression: May occur with concomitant CNS depressants or in the setting of underlying respiratory impairment
- Dizziness and somnolence: May impair patient's ability to drive or operate machinery
- Seizure frequency: May increase if rapidly discontinued
- Peripheral edema: Caution when coadministering with thiazolidinedione antidiabetic agents
Use in Special Populations
- Children and adolescents: Safety and effectiveness not established for various indications
Overdose Effects
Signs and symptoms include reduced consciousness, depression/anxiety, confusional state, agitation, restlessness, seizures, and heart block
