Indications

  • Neuropathic pain associated with diabetic peripheral neuropathy (DPN)
  • Postherpetic neuralgia (PHN)
  • Adjunctive therapy for the treatment of partial-onset seizures in patients 1 month of age and older
  • Fibromyalgia
  • Neuropathic pain associated with spinal cord injury

Pharmacology

  • Structural derivative of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA)
  • Binds with high affinity to the alpha2-delta site in central nervous system tissues

Dosage

  • Maximum recommended dose of 100 mg three times a day for patients with creatinine clearance of at least 60 ml/min
  • Recommended dose of 75 to 150 mg two times a day for postherpetic neuralgia in adults
  • Maximum recommended dose of 660 mg once daily for fibromyalgia in adults
  • Recommended dose range of 150 to 600 mg/day for neuropathic pain associated with spinal cord injury in adults
  • Conversion from Pregabalin capsules to Pregabalin CR capsule tablet

Administration

  • Oral route of administration
  • Should be swallowed whole and should not be split, crushed or chewed
  • Administered after an evening meal
  • Gradual tapering over a minimum of 1 week when discontinuing

Interaction

  • Unlikely to be involved in significant pharmacokinetic drug interactions

Contraindications

  • Contraindicated in patients with known hypersensitivity to Pregabalin or any of its components

  • Breastfeeding not recommended during treatment with pregabalin

  • Angioedema
  • Hypersensitivity reactions
  • Increased risk of suicidal thoughts or behavior
  • Respiratory depression
  • Dizziness and somnolence affecting driving or operating machinery
  • Increased seizure frequency or other adverse reactions if rapidly discontinued
  • Peripheral edema
  • Cautious coadministration with thiazolidinedione antidiabetic agents

Use in Special Populations

  • Safety and effectiveness not established in pediatric patients for certain indications
  • Safety and effectiveness not established for pediatric patients below the age of 1 month for adjunctive therapy for partial onset seizures
  • Safety and effectiveness of Pregaba extended-release tablet not established in pediatric patients

  • Confusional state
  • Agitation and restlessness
  • Seizures
