Description

  • Pregadel 50 mg Capsule is an anticonvulsant and is considered to be the first-line of treatment for neuropathic pain, shingles, seizures, and fibromyalgia.

Dosage and administration

  • Maximum recommended dose of Pregabalin is 100 mg three times a day for adults with diabetic peripheral neuropathy
  • Recommended dose of Pregabalin is 75 to 150 mg two times a day for postherpetic neuralgia in adults
  • Recommended dose of Pregabalin is 300 to 450 mg/day for fibromyalgia management in adults
  • Recommended dose range of Pregabalin is 150 to 600 mg/day for neuropathic pain associated with spinal cord injury in adults

  • Route of administration: Oral, with or without food
  • Pregabalin CR tablet should be administered after an evening meal

Pharmacology

  • Structural derivative of inhibitory neurotransmitter gamma-aminobutyric acid (GABA)
  • Binds with high affinity to the alpha2-delta site in central nervous system tissues

Contraindications

  • Contraindicated in patients with known hypersensitivity to Pregabalin or any of its components

Side effects

  • Most common side effects in adults: dizziness, somnolence, dry mouth, edema, blurred vision, weight gain, thinking abnormal
  • Most common side effects in pediatric patients: increased weight, increased appetite

Pregnancy and lactation

  • No adequate and well-controlled studies with pregabalin in pregnant women
  • Small amounts of pregabalin detected in the milk of lactating women
  • Breastfeeding not recommended due to potential risk of tumorigenicity

Precautions and warnings

  • Potential occurrence of angioedema requiring immediate discontinuation
  • Discontinuation if hypersensitivity reactions occur
  • Increased risk of suicidal thoughts or behavior
  • Monitoring and dosage adjustment for respiratory depression
  • Advice against driving or operating machinery
  • Caution with coadministration of Pregadel and thiazolidinedione antidiabetic agents
