Indications
- Neuropathic pain associated with diabetic peripheral neuropathy (DPN)
- Postherpetic neuralgia (PHN)
- Adjunctive therapy for the treatment of partial-onset seizures in patients 1 month of age and older
- Fibromyalgia
- Neuropathic pain associated with spinal cord injury
Pharmacology
Pregabalin is a structural derivative of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA). It binds with high affinity to the alpha2-delta site in central nervous system tissues, which may be involved in its effects on pain and seizures.
Dosage
Dosage varies based on the specific condition being treated, with different recommended starting doses and maximum doses for different indications.
Administration
- Route of administration: Oral
- Instructions: Pregabalin CR tablet should be swallowed whole after an evening meal and should not be split, crushed or chewed. When discontinuing, it should be gradually tapered over a minimum of 1 week.
Drug interaction
Unlikely to have significant pharmacokinetic drug interactions.
Contraindications
Contraindicated in patients with known hypersensitivity to Pregabalin or any of its components.

Precautions & Warnings
- Angioedema may occur and should be immediately addressed
- Discontinue immediately in case of hypersensitivity reactions
- Increased risk of suicidal thoughts or behavior
- May cause respiratory depression when used with concomitant CNS depressants or in the setting of underlying respiratory impairment
- May cause dizziness and somnolence
- Increased seizure frequency or other adverse reactions may occur if rapidly discontinued
- May cause peripheral edema
- Exercise caution when coadministering with thiazolidinedione antidiabetic agents
Use in Special Populations
- Children and adolescents: Safety and effectiveness not established for certain indications and age groups
Overdose Effects
Signs and symptoms include reduced consciousness, depression/anxiety, confusional state, agitation, restlessness, seizures, and heart block. There is no specific antidote.
